No for this workflow. Cross-store lines need a method such as Pickup, Delivery, or Warehouse Pickup, because the product is not sitting in your store for walk-out. Fulfilment usually flips away from Cash & Carry when stock comes from another outlet.

Yes. Separate from sale-driven ITOs, staff with Allow Transfers permission can send and receive stock from POS Transfers. That is for moving inventory between stores without ringing a customer sale. See stock transfers.
Not always. In Retail Express you can set Source to the stocking outlet and Fulfil to the store where the customer will pick up, or use Delivery so the goods go to them. The right Source/Fulfil pair drives whether an ITO is created.
